Uncertain significance for Cryopyrin associated periodic syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001243133.2(NLRP3):c.864C>G (p.Ile288Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 290 of the NLRP3 protein (p.Ile290Met). This variant is present in population databases (rs200900704, gnomAD 0.003%). This missense change has been observed in individual(s) with NLRP3-related conditions (PMID: 23432807, 33329557). This variant is also known as c.864C>G; p.I288M. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on NLRP3 protein function. Experimental studies have shown that this missense change affects NLRP3 function (PMID: 27548431).
