This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 4570, where C is replaced by T; at the protein level this means replaces leucine at residue 1524 with phenylalanine — a missense variant. Submitter rationale: The p.L1524F variant (also known as c.4570C>T), located in coding exon 29 of the ATM gene, results from a C to T substitution at nucleotide position 4570. The leucine at codon 1524 is replaced by phenylalanine, an amino acid with highly similar properties. In a study of whole-exome sequencing in patients with features of Cowden syndrome (CS) or Bannayan-Riley-Ruvalcaba syndrome (BRRS) and negative PTEN testing, this alteration was identified in 0/87 patients with CS or BRRS and 1/3476 patients from The Cancer Genome Atlas (TCGA) (Yehia L et al.
