Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004168.4(SDHA):c.1196C>A (p.Pro399Gln), citing Sema4 Curation Guidelines. This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 1196, where C is replaced by A; at the protein level this means replaces proline at residue 399 with glutamine — a missense variant. Submitter rationale: The SDHA c.1196C>A (p.P399Q) variant has not been reported in the literature to our knowledge. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 480869). Functional studies have not been performed and in silico tool predictions of the variants effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
