This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 457, where A is replaced by G; at the protein level this means replaces isoleucine at residue 153 with valine — a missense variant. Submitter rationale: The p.I153V variant (also known as c.457A>G), located in coding exon 5 of the SDHB gene, results from an A to G substitution at nucleotide position 457. The isoleucine at codon 153 is replaced by valine, an amino acid with highly similar properties. This alteration was observed in one individual in a cohort of 6238 PCC patients (Garrett A et al. Genet Med, 2022 Jan;24:41-50). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
