NM_003000.3(SDHB):c.550T>C (p.Tyr184His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 550, where T is replaced by C; at the protein level this means replaces tyrosine at residue 184 with histidine — a missense variant. Submitter rationale: The p.Y184H variant (also known as c.550T>C), located in coding exon 6 of the SDHB gene, results from a T to C substitution at nucleotide position 550. The tyrosine at codon 184 is replaced by histidine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.01% (greater than 10000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
