NM_003000.3(SDHB):c.308T>G (p.Met103Arg) was classified as Likely pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M103R variant (also known as c.308T>G), located in coding exon 4 of the SDHB gene, results from a T to G substitution at nucleotide position 308. The methionine at codon 103 is replaced by arginine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with SDHB-related hereditary pheochromocytoma-paraganglioma (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic.
