Uncertain significance for Gorlin syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003738.5(PTCH2):c.1205A>C (p.Tyr402Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with serine, which is neutral and polar, at codon 402 of the PTCH2 protein (p.Tyr402Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PTCH2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on PTCH2 protein function.
