Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000163.5(GHR):c.3G>A (p.Met1Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change affects the initiator codon of the GHR mRNA. This change may impact translation initiation or efficiency. The next in-frame methionine is located at codon 188. This variant is not present in population databases (gnomAD no frequency). Disruption of the initiator codon has been observed in individuals with autosomal recessive Lardon syndrome (PMID: 12199334, 29025428, 31429861). It has also been observed to segregate with disease in related individuals. This variant disrupts a region of the GHR protein in which other variant(s) (p.Glu62Lys) have been observed in individuals with GHR-related conditions (PMID: 7565946). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
