Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002878.4(RAD51D):c.877G>A (p.Ala293Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the RAD51D gene (transcript NM_002878.4) at coding-DNA position 877, where G is replaced by A; at the protein level this means replaces alanine at residue 293 with threonine — a missense variant. Submitter rationale: The p.A293T variant (also known as c.877G>A), located in coding exon 9 of the RAD51D gene, results from a G to A substitution at nucleotide position 877. The alanine at codon 293 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 175000 alleles tested) in our clinical cohort. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
