This variant lies in the ALG13 gene (transcript NM_001099922.3) at coding-DNA position 40, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 14 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 14 of the ALG13 protein (p.Asp14Asn). This variant is present in population databases (rs139688782, gnomAD 0.004%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with ALG13-related conditions.
