This variant lies in the AKAP9 gene (transcript NM_005751.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 6331, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 26 of the AKAP9 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in AKAP9 cause disease.
