NM_005732.4(RAD50):c.3538C>T (p.Arg1180Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1180W variant (also known as c.3538C>T), located in coding exon 23 of the RAD50 gene, results from a C to T substitution at nucleotide position 3538. The arginine at codon 1180 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
