This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 146, where A is replaced by G; at the protein level this means replaces asparagine at residue 49 with serine — a missense variant. Submitter rationale: This missense variant replaces asparagine with serine at codon 49 of the PTEN protein. Computational prediction tool is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). A high throughput functional study showed a neutral impact on lipid phosphatase activity (PMID: 29706350). This variant has not been reported in individuals affected with PTEN-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
