Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.1001T>C (p.Ile334Thr), citing Ambry Variant Classification Scheme 2023: The p.I334T variant (also known as c.1001T>C), located in coding exon 10 of the PMS2 gene, results from a T to C substitution at nucleotide position 1001. The isoleucine at codon 334 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
