Uncertain significance for Galactosylceramide beta-galactosidase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000153.4(GALC):c.158G>A (p.Arg53Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 53 of the GALC protein (p.Arg53Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with a positive newborn screening result for GALC-related disease (PMID: 27638593). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on GALC protein function. Experimental studies have shown that this missense change does not substantially affect GALC function (PMID: 27638593). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
