This variant lies in the MED12 gene (transcript NM_005120.3) at coding-DNA position 3469, where A is replaced by G; at the protein level this means replaces asparagine at residue 1157 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces asparagine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 1157 of the MED12 protein (p.Asn1157Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Opitz-Kaveggia syndrome (PMID: 36271811). In at least one individual the variant was observed to be de novo. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt MED12 protein function with a negative predictive value of 95%.
