Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.44T>C (p.Leu15Pro), citing Ambry Variant Classification Scheme 2023: The p.L15P variant (also known as c.44T>C), located in coding exon 1 of the ATM gene, results from a T to C substitution at nucleotide position 44. The leucine at codon 15 is replaced by proline, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally abnormal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
