NM_000179.3(MSH6):c.875G>C (p.Ser292Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S292T variant (also known as c.875G>C), located in coding exon 4 of the MSH6 gene, results from a G to C substitution at nucleotide position 875. The serine at codon 292 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. In addition, the CoDP in silico tool predicts this alteration to have minor impact on molecular function, with a score of 0.000 (Terui H et al. J. Biomed. Sci. 2013;20:25).
