Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005591.4(MRE11):c.524A>G (p.Lys175Arg), citing Ambry Variant Classification Scheme 2023: The p.K175R variant (also known as c.524A>G), located in coding exon 5 of the MRE11A gene, results from an A to G substitution at nucleotide position 524. The lysine at codon 175 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
