Uncertain significance for Ataxia-telangiectasia-like disorder — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005591.4(MRE11):c.1205G>C (p.Arg402Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with threonine at codon 402 of the MRE11 protein (p.Arg402Thr). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and threonine. This variant is present in population databases (rs778211645, ExAC 0.009%). This variant has not been reported in the literature in individuals with MRE11-related conditions. ClinVar contains an entry for this variant (Variation ID: 479718). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
