Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.603T>G (p.Phe201Leu), citing Ambry Variant Classification Scheme 2023: The p.F201L variant (also known as c.603T>G), located in coding exon 4 of the CHEK2 gene, results from a T to G substitution at nucleotide position 603. The phenylalanine at codon 201 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
