NM_007194.4(CHEK2):c.275C>A (p.Pro92His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P92H variant (also known as c.275C>A), located in coding exon 1 of the CHEK2 gene, results from a C to A substitution at nucleotide position 275. The proline at codon 92 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
