Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.88G>A (p.Gly30Ser), citing Ambry Variant Classification Scheme 2023: The p.G30S variant (also known as c.88G>A), located in coding exon 1 of the CHEK2 gene, results from a G to A substitution at nucleotide position 88. The glycine at codon 30 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
