This variant lies in the CACNA1H gene (transcript NM_021098.3) at coding-DNA position 1687, where G is replaced by C; at the protein level this means replaces glycine at residue 563 with arginine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 563 of the CACNA1H protein (p.Gly563Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with idiopathic trigeminal Neuralgia (PMID: 33083721). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CACNA1H protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change affects CACNA1H function (PMID: 35467587).
