This variant lies in the NPR2 gene (transcript NM_003995.4) at 3 bases into the intron immediately after coding-DNA position 2047, where G is replaced by A. Submitter rationale: This sequence change falls in intron 13 of the NPR2 gene. It does not directly change the encoded amino acid sequence of the NPR2 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs200461596, gnomAD 0.05%). This variant has not been reported in the literature in individuals affected with NPR2-related conditions. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
