NM_000059.4(BRCA2):c.8158G>A (p.Glu2720Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2720K variant (also known as c.8158G>A), located in coding exon 17 of the BRCA2 gene, results from a G to A substitution at nucleotide position 8158. The glutamic acid at codon 2720 is replaced by lysine, an amino acid with similar properties. This alteration has been reported in a cohort of individuals with prostate cancer who underwent testing for alterations in BRCA2; authors listed p.E2720K as an unclassified variant in this study (Kote-Jarai Z et al. Br. J. Cancer, 2011 Oct;105:1230-4). This amino acid position is well conserved through mammals but not in all available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
