Uncertain significance for Idiopathic generalized epilepsy; Hyperaldosteronism, familial, type IV — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021098.3(CACNA1H):c.2908-17_2908-11del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CACNA1H gene (transcript NM_021098.3) at 17 bases into the intron immediately before coding-DNA position 2908 through 11 bases into the intron immediately before coding-DNA position 2908, deleting this region. Submitter rationale: This sequence change falls in intron 13 of the CACNA1H gene. It does not directly change the encoded amino acid sequence of the CACNA1H protein. This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with CACNA1H-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
