Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000059.4(BRCA2):c.8119A>G (p.Lys2707Glu), citing Sema4 Curation Guidelines. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 8119, where A is replaced by G; at the protein level this means replaces lysine at residue 2707 with glutamic acid — a missense variant. Submitter rationale: The BRCA2 c.8119A>G (p.K2707E) variant has not been reported in the literature to our knowledge. It was observed in 1/251356 chromosomes in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 479316). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
