This variant lies in the KIF1A gene (transcript NM_001244008.2) at coding-DNA position 5333, where G is replaced by T; at the protein level this means replaces arginine at residue 1778 with leucine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 1677 of the KIF1A protein (p.Arg1677Leu). This variant also falls at the last nucleotide of exon 45, which is part of the consensus splice site for this exon. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant has not been reported in the literature in individuals affected with KIF1A-related conditions.
