This variant lies in the NPR2 gene (transcript NM_003995.4) at coding-DNA position 3020, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 1007 with glycine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 1007 of the NPR2 protein (p.Asp1007Gly). This variant is present in population databases (rs768235885, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with NPR2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt NPR2 protein function with a negative predictive value of 80%.
