NM_000051.4(ATM):c.8609A>G (p.Asp2870Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2870G variant (also known as c.8609A>G), located in coding exon 58 of the ATM gene, results from an A to G substitution at nucleotide position 8609. The aspartic acid at codon 2870 is replaced by glycine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally abnormal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
