Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.1852A>G (p.Lys618Glu), citing Ambry Variant Classification Scheme 2023: The p.K618E variant (also known as c.1852A>G), located in coding exon 11 of the ATM gene, results from an A to G substitution at nucleotide position 1852. The lysine at codon 618 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
