This variant lies in the CHCHD10 gene (transcript NM_213720.3) at coding-DNA position 127, where G is replaced by T; at the protein level this means replaces glycine at residue 43 with tryptophan — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with tryptophan, which is neutral and slightly polar, at codon 43 of the CHCHD10 protein (p.Gly43Trp). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CHCHD10-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
