Uncertain significance for Epilepsy, childhood absence, susceptibility to, 1; Epilepsy, childhood absence, susceptibility to, 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000814.6(GABRB3):c.328A>C (p.Asn110His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with histidine, which is basic and polar, at codon 110 of the GABRB3 protein (p.Asn110His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GABRB3-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on GABRB3 protein function. This variant disrupts the p.Asn110 amino acid residue in GABRB3. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 23934111, 26950270). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
