NM_000051.4(ATM):c.6298T>C (p.Tyr2100His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y2100H variant (also known as c.6298T>C), located in coding exon 42 of the ATM gene, results from a T to C substitution at nucleotide position 6298. The tyrosine at codon 2100 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
