This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 1310, where C is replaced by A; at the protein level this means replaces proline at residue 437 with histidine — a missense variant. Submitter rationale: This sequence change replaces proline, which is neutral and non-polar, with histidine, which is basic and polar, at codon 437 of the SQSTM1 protein (p.Pro437His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with amyotrophic lateral sclerosis (PMID: 31859009). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
