This variant lies in the TCAP gene (transcript NM_003673.4) at coding-DNA position 44, where G is replaced by C; at the protein level this means replaces cysteine at residue 15 with serine — a missense variant. Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with serine, which is neutral and polar, at codon 15 of the TCAP protein (p.Cys15Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TCAP-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
