This variant lies in the TCAP gene (transcript NM_003673.4) at coding-DNA position 449, duplicating one base; at the protein level this means shifts the reading frame starting at alanine residue 151, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the TCAP protein (p.Ala151Cysfs*58). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 17 amino acid(s) of the TCAP protein and extend the protein by 40 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TCAP-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
