This variant lies in the TNNT2 gene (transcript NM_001276345.2) at coding-DNA position 869, deleting one base; at the protein level this means shifts the reading frame starting at lysine residue 290, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the TNNT2 protein (p.Lys280Argfs*68). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 9 amino acid(s) of the TNNT2 protein and extend the protein by 58 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TNNT2-related conditions.
