Pathogenic for Leber congenital amaurosis 2; Retinitis pigmentosa 20 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000329.3(RPE65):c.1243+2T>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 11 of the RPE65 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or altered protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with Leber congenital amaurosis (PMID: 25383945). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Studies have shown that disruption of this splice site results in activation of a cryptic splice site, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 25383945). For these reasons, this variant has been classified as Pathogenic.
