This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 180, where G is replaced by C; at the protein level this means replaces lysine at residue 60 with asparagine — a missense variant. Submitter rationale: This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 60 of the BAG3 protein (p.Lys60Asn). This variant also falls at the last nucleotide of exon 1, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BAG3-related conditions.
