This variant lies in the MITF gene (transcript NM_001354604.2) at coding-DNA position 1552, deleting one base; at the protein level this means shifts the reading frame starting at serine residue 518, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the MITF protein (p.Ser411Alafs*25). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 9 amino acid(s) of the MITF protein and extend the protein by 15 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MITF-related conditions.
