Submitter rationale: This sequence change inserts a large fragment of DNA, likely a transposable element, in exon 339 of the TTN gene (c.94142_94143ins?), causing a frameshift at codon 31381 (p.Phe31381fs). The exact size and sequence of the insertion cannot be determined by the current assay. While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. This variant is not present in population databases (gnomAD no frequency). A similar variant has been observed in individual(s) with dilated cardiomyopathy (internal data). This variant is located in the A band of TTN (PMID: 25589632). Truncating variants in this region are significantly overrepresented in patients affected with dilated cardiomyopathy (PMID: 25589632).
