Uncertain significance for Mevalonic aciduria; Hyperimmunoglobulin D with periodic fever; Porokeratosis 3, disseminated superficial actinic type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000431.4(MVK):c.839_883del (p.Gly280_Leu294del), citing Invitae Variant Classification Sherloc (09022015): This variant, c.839_883del, results in the deletion of 15 amino acid(s) of the MVK protein (p.Gly280_Leu294del), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MVK-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant disrupts a region of the MVK protein in which other variant(s) (p.Met282Thr) have been observed in individuals with MVK-related conditions (PMID: 28603204). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
