This variant lies in the BAG3 gene (transcript NM_004281.4) at coding-DNA position 1429 through coding-DNA position 1430, inserting TG; at the protein level this means shifts the reading frame starting at arginine residue 477, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Arg477Leufs*90) in the BAG3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 99 amino acid(s) of the BAG3 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BAG3-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
