This variant lies in the CASR gene (transcript NM_000388.4) at coding-DNA position 2333 through coding-DNA position 2339, deleting 7 bases; at the protein level this means shifts the reading frame starting at glycine residue 778, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Gly778Alafs*57) in the CASR gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 301 amino acid(s) of the CASR protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CASR-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
