Pathogenic for Paget disease of bone 2, early-onset; Frontotemporal dementia and/or amyotrophic lateral sclerosis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003900.5(SQSTM1):c.838G>T (p.Glu280Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 838, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 280 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu280*) in the SQSTM1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SQSTM1 are known to be pathogenic (PMID: 27545679, 29959261). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with SQSTM1-related condition (PMID: 37205252). For these reasons, this variant has been classified as Pathogenic.
