NM_000248.4(MITF):c.20A>G (p.Tyr7Cys) was classified as Uncertain significance for Melanoma, cutaneous malignant, susceptibility to, 8; Waardenburg syndrome type 2A; Tietz syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 7 of the MITF protein (p.Tyr7Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant Waardenburg syndrome (PMID: 24194866). It has also been observed to segregate with disease in related individuals. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
