This variant lies in the RPE65 gene (transcript NM_000329.3) at coding-DNA position 1499, where C is replaced by A; at the protein level this means replaces alanine at residue 500 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 500 of the RPE65 protein (p.Ala500Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive Leber congenital amaurosis (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
