This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 625, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 209 with lysine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with lysine at codon 209 of the RYR1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with malignant hyperthermia susceptibility, this individual also carried a known pathogenic variant in the RYR1 gene that could explain the observed phenotype (PMID: 21455645). This variant has been identified in 5/248998 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
